Optimasi metode isolasi RNA dari jaringan kanker payudara fiksasi formalin dengan nucleospin RNA II kit macherey-nagel / Lulus Putri Aninda
Abstrak
Breast cancer is a serious public health problem therefore an identification of genetic factors which plays role in cancer development needed to thoroughly as an effort how to prevent the disease. Research based on RNA from formalin-fixed tissue is an important for clinic and molecular analysis. Fixation method and extraction on its sample generally can obstruct nucleic acid acquisition from sample. This research was conducted to optimize the method RNA extraction from breast cancer formalin-fixed tissue to obtain the pure and high quantity of total RNA. The result shown RNA extraction yield using modified protocol NucleoSpin RNA II Macherey-Nagel kit were have higher total RNA quantify than original protocol. Modification was conducted at buffer usage to wash formalin-fixed tissue crushing the tissue cell lysis stage and RNA binding condition adjustment. The next optimization is needed to avoid contamination.
